产MBLs的牛蛙源ST520型维氏气单胞菌的致病性和耐药性分析

Analysis of pathogenicity and drug resistance of ST520 Aeromonas veronii from Lithobates catesbeiana producing MBLs

    目的 探究湖北省荆州市某牛蛙养殖场患病牛蛙皮肤出血、腹水症状的病因。
    方法 本实验从患病牛蛙肠组织中分离一株优势菌,通过形态学观察、生理生化鉴定、16S rRNA基因测序,结合MLST分析、毒力基因检测、致病性实验、耐药基因检测和药物敏感性实验等方法分析其致病性和耐药性。
    结果 分离菌株为革兰氏阴性菌,16S rRNA基因序列比对显示,分离菌株与维氏气单胞菌(MK584925.1)序列相似度最高,同源性达99.90%,确定该菌株为牛蛙源维氏气单胞菌,命名为W0525。多位点序列分型(MLST)确定分离菌株为ST520型;该菌株携带5种毒力基因(aer、act、alt、ahyBexu),具有β溶血活性;人工感染实验发现死亡牛蛙皮肤红肿、腹腔出现腹水、胃肠道出现炎症病变,与自然感染临床症状相似;分离菌株感染牛蛙的半数致死量(LD50)为5.62×108 CFU/mL,其中肠组织的载菌量最高。分离菌株携带5种耐药基因(bla-TEM、bla-CphA、aadA1、aadA2和tetA),对青霉素、阿莫西林等16种抗生素耐药,具有强生物被膜形成能力,且对中药苏木高度敏感。分离菌株W0525为产MBLs的多重耐药菌株,其bla-CphA基因与bla-CphA4聚为一支。
    结论 本研究从牛蛙病变组织中成功分离到一株维氏气单胞菌,并对致病性、耐药性等方面进行研究。研究结果为牛蛙源维氏气单胞菌的感染和精准施治提供理论依据和数据支撑。

     

    Abstract: Lithobates catesbeiana (American bullfrog) is extensively cultivated in China, valued by consumers for its tender texture, palatable taste, and abundant nutritional components.This study aimed to identify the etiological agent causing skin hemorrhage and ascites in diseased L. catesbeiana collected from a bullfrog farm in Jingzhou, Hubei Province. A dominant bacterium was isolated from the intestinal tissue of diseased L. catesbeiana. Its pathogenicity and drug resistance were analyzed through morphological observation, physiological and biochemical identification, 16S rRNA gene sequencing, combined with MLST analysis, virulence gene detection, pathogenicity experiments, drug resistance gene detection, and drug susceptibility testing. The isolated strain was a Gram-negative bacterium, and 16S rRNA gene sequence alignment revealed highest similarity with A. veronii (MK584925.1), confirming it as A. veronii isolated from L. catesbeiana and designated W0525. MLST analysis determined that the isolated strain was of the ST520 type. The strain carried five virulence genes: aer, act, alt, ahyB, and exu, and exhibited β-hemolytic activity. In the artificial infection experiments, the deceased L. catesbeiana showed clinical signs similar to natural infection, including skin redness and swelling, abdominal ascites, and gastrointestinal inflammatory lesions. The median lethal dose (LD50) of the isolate to L. catesbeiana was 5.62×108 CFU/mL. The isolate carried five resistance genes: bla-TEM, bla-CphA, aadA1, aadA2, and tetA. It demonstrated strong biofilm-forming ability and was resistant to 16 antibiotics, including penicillin and amoxicillin, but highly sensitive to the traditional Chinese medicine Caesalpinia sappan (Sappan wood). Strain W0525 was identified as a metallo-β-lactamase (MBL)-producing multidrug-resistant strain, and its bla-CphA gene clustered with bla-CphA4 in phylogenetic analysis. This study successfully isolated and characterized an MDR A. veronii strain from diseased L. catesbeiana, providing critical insights into its pathogenic mechanisms and antimicrobial resistance profile. These findings establish a theoretical foundation and empirical data for the development of targeted prevention and control strategies against A. veronii infections in bullfrog aquaculture.
